چکیده

The livers of rats fed 10 diets previously found to inhibit or accelerate the induction of tumors by 4-dimethylaminoazobenzene were analyzed for riboflavin, biotin, and vitamin B(6) after 6 and 19 weeks on the diets. Those fed diets accelerating tumor induction had average hepatic riboflavin levels of 9 to 11 microg. per gm. The livers of the control and protected rats averaged 14 and 15 to 19 microg. per gm., respectively. The hepatic levels of vitamin B(6) and biotin averaged 7.0 and 0.54 microg. per gm., respectively, and were independent of the protective nature of the diet. The livers of rats fed the same diets in the absence of 4-dimethylaminoazobenzene contained about 50 per cent more of each of these vitamins than those from dye-fed rats. In a second series the tumor incidence and the hepatic riboflavin and vitamin B(6) were determined simultaneously. In this experiment urethane decreased the level of hepatic riboflavin and increased the rate of induction of tumors slightly. Methionine inhibited tumor induction to a small extent while cystine had a more pronounced retarding effect; the level of hepatic riboflavin for both groups was higher than in the controls. The livers of rats fed the basal diet, 0.054 per cent of 4-dimethylaminoazobenzene, or equimolar amounts of its 2-, 2'-, 3'-, or 4'-methyl derivatives were analyzed for riboflavin at various times over a 16 week period. In each case the level of hepatic riboflavin decreased throughout the experimental period, but the rate and extent of the riboflavin loss were greatest with the more carcinogenic compounds.

those of the parent compound, and the degree of change was approximately proportional to the carcinogenic potency of the derivative fed. In the present paper these comparisons are ex tended to the livers of two species, the mouse and the golden hamster, which are highly resistant to the carcinogenic action of 4-dimethylaminoazo benzene. With this agent mice develop tumors only slowly (5, 15) an...

Studies on the intracellular composition of liver and liver tumor from rats fed 4-dimethylaminoazobenzene.

In a previous study (8) the livers of rats fed the hepatic carcinogen, 4-dimethylaminoazobenzene, were separated by differential centrifugation into the nuclei, large granules (mitochondria), small granules (microsomes), and a supernatant fluid (particles nonsedimentable at 19,000 X g). Anal yses for protein, nucleic acids, riboflavin, and pro tein-bound aminoazo dye were made on each frac tion...

Studies on the intracellular composition of livers from rats fed 2-acetylaminofluorene.

Price and his associates (17, 18, 19) found al terations in the amounts of protein, nucleic acids, and riboflavin in various fractions of the liver during the induction of liver tumors by a series of aminoazo dyes of widely different carcinogenic ac tivities. The changes induced by the more active carcinogens made the composition of the liver ap proach that of the tumors induced by the parent c...

Hepatic Riboflavin and Tumor Formation in Rats Fed Azo Dyes in Various Diets*

I t is well recognized that rats fed p-dimethylaminoazobenzene (DAB) develop tumors at rates that depend upon the composition of the basal diet. Dietary factors reported to increase the rate of tumor formation under certain circumstances include biotin (2, 4), pyridoxine (16), cystine (18), and the rice bran concentrate "Vitab" (12, 13, 16). The dietary factors most effective in retarding tumor...

The in vitro formation of protein-bound derivatives of aminoazo dyes by rat liver preparations.

Previous studies have demonstrated the presence of protein-bound aminoazo dyes in the livers of rats fed the hepatocarcinogen 4-dimethylaminoazobenzene (DAB) 1 or certain of its derivatives ( ~ , 35). Since, under a variety of conditions, the levels of these bound derivatives can be correlated with the expected tumor incidence, the formation of the protein-bound dyes appears to be an important ...
